How it works

Oxytocin is a cyclic 9-amino-acid peptide (Cys-Tyr-Ile-Gln-Asn-Cys-Pro-Leu-Gly-NH2, molecular formula C43H66N12O12S2, ~1,007 daltons). The two cysteine residues form a sulfur bridge that gives the molecule a small ring plus a short tail.

It is made in the paraventricular and supraoptic nuclei of the hypothalamus and released two ways. Released into the bloodstream from the posterior pituitary, it acts on uterine smooth muscle to drive labor contractions and on mammary myoepithelial cells to trigger milk let-down. Released centrally from oxytocin neurons that project to other brain regions, it acts on the oxytocin receptor (a G-protein-coupled receptor) in places like the amygdala, nucleus accumbens, and brainstem.

The behavioral effects studied in human research - changes in social attention, trust, and emotional response to faces - depend on central oxytocin activity. That is the main reason peripheral routes like subcutaneous injection are not expected to reproduce intranasal effects: the molecule does not cross the blood-brain barrier easily, and only a small fraction of intranasally administered oxytocin (about 1-2%) reaches the bloodstream, with a smaller fraction reaching the brain through nose-to-brain pathways.
